A rare natural chiral amino acid derivative, a crystalline sulfur-containing compound originally isolated from ergot fungi. In the dissolved state, ergothione exists as a tautomer between its thione and thiol structures. At physiological pH, ergothionein is mainly in the form of thione, so it will only react with some sulfhydryl reagents.

The operation of ergothioneine in organisms depends on a special transporter OCTN1, which exists in keratinocytes and melanocytes and has a high affinity for ergothione. Therefore, ergothioneine is easily absorbed and fully utilized in skin tissues and cells. Through OCTN1 in cell membrane and mitochondrial membrane, ergothioneine is transported to mitochondria, the main "focus" of free radical generation in cells, which can protect mitochondria from oxidative damage while directly removing reactive oxygen free radicals, so as to achieve the ultimate goal of delaying the aging of human cells.

It can be seen from the experimental results that ergothioneine has better free radical scavenging effect. With the increase of concentration, the free radical scavenging effect of ergothioneine is more significant(Fig. a). After UV irradiation, the content of intracellular ROS increased significantly, which was 108% higher than that of normal cells. In the ergothioneine group produced by Readline, the amount of ROS was significantly reduced, with increase of concentration, the amount of ROS decreased significantly. From the experimental results, it can be seen that ergothioneine has a significant effect of scavenging intracellular ROS, indicating that ergothioneine has excellent antioxidant effect.(Fig. b)
